A Whole-Genome Sequence Genome-Wide Association Study in All of Us Identifies a Novel Glaucoma Risk Locus in African Individuals.

While primary open-angle glaucoma (POAG) is prevalent in all populations, population-based studies suggest it disproportionately affects African and Latino individuals, who have a younger age at onset, present at more advanced disease stages, and have greater risk of blindness compared to non-Hispanic Whites.1, 2, 3, 4, 5 More than 300 risk loci have been identified in prior analyses of this highly heritable complex disease,6, 7, 8 which have mainly been conducted in individuals of European (EUR) ancestry. However, few are conclusively associated with risk of glaucoma in individuals of African (AFR) continental genetic ancestry,9, 10, 11, 12, 13, 14 and even fewer in those with Latino/Admixed American (AMR) ancestry.10,14
